polycloning

The oligonucleotides were annealed and cloned into the pmirGLO vector (Promega Corporation, USA) between the XhoI/XbaI restriction sites of the polycloning site (PCS), resulting in the miRNA target region in the correct 5´ to 3´ orientation and immediately downstream of the luciferase gene.
